Shanghai Pasteur developed for the intestinal virus EV71 neutralizing monoclonal

Recently, the international academic journals, Journal of Virological Methods, published online Chinese Academy of Sciences, Institut Pasteur of Shanghai Huang research group successfully prepared for the latest research results of the intestinal virus EV71 neutralizing monoclonal antibody.
The intestinal virus EV71 cause hand, foot and mouth disease pathogens. HFMD often occurs in children under the age of five infectious disease, some patients with severe neurological symptoms of encephalitis, pulmonary edema and paralysis or even death. In recent years, hand, foot and mouth disease in the Asia-Pacific region, widespread and poses a serious threat to the healthy development of children. In China, only the 2010 and 2011 the Ministry of Health statistics, on a total of 3,394,375 cases of infection, of which 1414 cases of death. However, the lack of simple and rapid detection of EV71 virus, and no preventive vaccines and therapeutic drugs. Therefore, the development of diagnostic reagents and antivirals is of great significance for the prevention and treatment of foot and mouth disease.
Pasteur of Shanghai, PhD library Zhiqiang Shi Jinping and Liu Qingwei in Huang researcher under the guidance of, the use of genetic engineering methods to prepare infectious EV71 virus-like particles as immunogens, with anti-immunized mice to produce virus ability of neutralizing antibodies. Then, using traditional hybridoma technology preparation of the three that secrete neutralizing antibody hybridoma D5, C4 and H7.
The biochemical analysis showed that these monoclonal antibodies can specifically recognize the surface of the capsid protein of EV71 virus. Immunofluorescence experiments showed that these antibodies are also able to detect EV71 virus infected cells. Therefore, these monoclonal antibodies can be used to further the development of diagnostic kits for EV71. More importantly, in vitro experiments, the monoclonal antibody in very low concentrations can effectively prevent the virus from invading cells, suggesting that they have developed into a humanized monoclonal antibody drug for the treatment of the potential.

The study of early diagnosis of EV71 infection and monoclonal antibody drug development has laid a good foundation. The work was supported by the Chinese Academy of Sciences "Hundred Talents Program" funded.
